A multitude of stimuli influences the production of reactive oxygen species (ROS). The family of NADPH oxidases (Nox) is one of the most important sources of ROS. Nox4 constitutively produces ROS whereas all other Nox-proteins need to be activated by agonists. The physiological role of Nox4, its mechanism of ROS-production and its relevance for cellular signal transduction so far are unidentified and will be analyzed in this subproject. Our analyses will focus on both: the enzymatic function of Nox4 as well as on the physiological and pathophysiological role of Nox4-generated ROS.
